Roles of Chromatin insulators in gene regulation and diseases

With advances in genetic science, the dynamic structure of eukaryotic genome is considered as basis of gene expression regulation. Long-distance communication between regulatory elements and target promoters is critical and the mechanisms responsible for this connection are just starting to emerge. Chromatin insulators are key determinants of proper gene regulation and precise organization of chromosomal structures. These elements are DNA-protein complexes which can mediate interactions within and between chromosomes. In doing so, they can bring close together regulatory sequences located at large distances from each other. In addition, insulators may be important in remodeling the architecture of chromosomes in the cell cycle. Here, we review recent data revealing the structure and properties of these DNA-protein complexes, their global role in regulation of transcription, and the mechanisms by which misregulation of their function leads to various human diseases such as cancers and autoimmune diseases.

Minimal HER1 and HER2 expressions in CHO and HEK-293 cells cause them appropriate negative cells for HERs-related studies

Background: Understanding of cellular and molecular mechanisms involved in cell/tumor growth and progression has led to molecular-targeted therapy. HER1 and HER2 are two main oncogenic components of HER-receptor family considered as targets for cancer research and therapy. Reliable results of HER- related research and targeted therapies dependent upon cell line with known HER expression status. But there are no published studies focusing on determination of HER1 and HER2 receptors expression in CHO and HEK-293 cell lines.
Materials and Methods: Absolute expressions of HER1 and HER2 in CHO, HEK-293, MDA-MB-468 and SKBR3 cells were evaluated using quantitative real-time PCR. Absolute expression quantification was carried out by serial dilution plasmids harboring HER1 and HER2 to extrapolate standard curve.
Results: Real-time PCR amplification was successfully optimized by HER1 and HER2 plasmids with high efficiency and their copy numbers were calculated. The results showed that HER1 was not expressed in CHO and HEK-293 cells but these cells express HER2 up to 9.4 × 102 and 1.1 × 105 copy/µg, respectively. MDA-MB-468 and SKBR3 cells express HER1 up to 8.4 × 105 and 9.7 × 104, respectively. In addition, the expression of HER2 is higher on the surface of SKBR3 cell (6.5 × 106 copy/µg).
Conclusion: Our study provides a proven insight to HER1 and HER2 expression status in CHO, HEK-293, MDA-MB-468 and SKBR3 cells. Heterologous expression of HER1 and HER2 in cell lines is the first step in structural and functional studies. Since HER1 is not expressed in CHO and HEK-293 endogenously, these cells are ideal for HER receptor related studies.

Evaluation of a new bombesin analogue labeled with 99mTc as potential targeted tumor scintigraphic agent

Background: Bombesin shows high affinity for Gastrin-releasing peptide (GRP) receptors which over expressed on the cell surfaces of several human tumors particularly in prostate and breast cancers. The aim of this study was labeling of designed analogue with99mTc via HYNIC and Tricine /EDDA and evaluation as potential targeted tumor scintigraphic agent.
Materials and Methods: HYNIC-Bombesin was prepared by solid phase synthesis using Fmoc strategy and radiolabeled with 99mTc at 100 °C for 10 min by exchange method and radiochemical analysis involved ITLC and HPLC methods. The stability of radiopeptide was checked in the presence of human serum at 37 °C up to 24 h. Internalization was studied with the human GRP receptor cell line PC-3. Biodistribution study was performed in mice.
Results: Radiochemical purities of >98% was obtained. Radiopeptide showed high stability in serum. Radioligand internalization into PC-3 cells was high and specific. Biodistribution study demonstrated that 99mTc-HYNIC peptide cleared fast from blood and most non-targeted tissues and was excreted mainly by renal pathway and was uptake significantly in GRPr positive tissues such as pancreas.
Conclusion: Easy radiolabeling of peptide conjugate together with favorable in vitro and in vivo characteristics might be a useful peptide radiopharmaceutical in diagnosis of GRPr positive tumors.

Background: Coronary artery disease (CAD) is one of the leading causes of death and disability around the world. Interaction between genetic and environmental factors determines susceptibility of an individual to develop coronary artery disease . Lipoprotein lipase (LPL) play an important role in the metabolism of HDL-C ( High Density Lipoprotein Cholesterol ), LDL-C (Low Density Lipoprotein Cholesterol ) and triglycerides (TG). Dysfunction of LPL as a result of genetic variants of lipoprotein lipase gene is associated with increased risk of CAD. The aim of the present study was to investigate the relationship between the risk of coronary artery disease and LDL-C, HDL-C and TG (triglycerides) levels by lipoprotein lipase gene Hind III polymorphism.
Materials and Methods: A total of 202 subjects including 114 patients with coronary artery disease and 88 control participated in this study. The Hind III polymorphism of the lipoprotein lipase gene was determined by PCR- RFLP (Polymerase Chain Reaction-Restriction Fragment Length Polymorphism) . In the presence and absence of restriction site, the genotypes are described H+/+ , H-/- respectively.
Results: In this survey, a highly significant association between the frequent H+/+ genotype and unfavorable TG levels was observed in our population . For the Hind III genotypes, within the healthy subjects (n=88), the H+/+ genotype was found in 67 individuals (58.8%), H-/+ genotype in 38 individuals (33.3%) , and 9 individuals (7.8%) carried the H-/- genotype. Within the CAD group (n=114), 47 individuals (53.4%) with H+/+ genotype, 36 (41%) with H-/+ genotype, and 5 (5.6%) carried the H-/- genotype.
Conclusion: There was a significant difference between the distribution of LPL–Hind III genotypes and the healthy subjects and the patients with CAD (P<0.05, 0. 645). LPL–Hind III polymorphisms were not detected as independent risk factors for CAD in this study group, but had significant associations with TG levels (P<0.05).

Epicatechin enhances anti-proliferative effect of bleomycin in ovarian cancer cell

Background: Bleomycin (BLM) is an anti-cancer drug widely used in the treatment of cancer. BLM causes several side effects related to DNA and cellular damage. The aim of this study was investigated the effects of tea polyphenol epicatechin on anti-proliferative effects induced by bleomycin in human normal skin and human ovarian cancer cells.
Materials and Methods: Human ovarian cancer cell (SKOV-3) and human non-malignant fibroblast cell (HFFF2) were treated with epicatechin at various concentrations (10, 25 and 50 µM) and BLM alone and with their combinations, further their effects on cell viability were evaluated.
Results: The combined treatment of epicatechin with BLM enhanced significantly inhibition of cell growth in comparison to BLM alone in cancer cell. Epicatechin enhanced significantly cytotoxicity induced by BLM with 83% at dose 50 µM, while it was 92% in BLM-treated cells. Epicatechin was not showed any cytotoxicity on HFFF2 cells.
Conclusion: Study suggests that epicatechin chemosensitize the ovarian cancer cell to BLM-induced growth inhibition without any toxicity on normal cell.

Effect of Teucrium polium leaf extracts on AMPK level in Isolated Rat Pancreases

Background: The purpose of this study is to evaluate effects of Teucrium polium (TP) leaf extracts on adenosine monophosphate activated protein kinase (AMPK) level in isolated pancreases.
Materials and Methods: In this experimental study, two groups of rats were used as a control group, and diabetic group. In the end of experimental period the animals were sacrificed then pancreas were removed, then isolated rat pancreas was cultured in buffer with or without TP leaf extracts. Measuring the sample glucose level was carried out by spectrophotometric method ( Jenway ,Model 6505 ,UK, Pars Azmmon Co. ,Tehran, IRAN) . Insulin level was assayed by an enzyme immunoassay ELISA kit specific for rats made by Mercodia Rat insulin ELISA ( 10-1250-01,Mercodia AB, Uppsala, Sweden). The adenosine monophosphate activated protein kinase level was determined by ELISA assay method using a rat phosphorylated AMPK ELISA kit ( CSB-E11337r, Cusabio Biotech Co .,LTD).
Results: The results indicate that addition of TP leaf extract significantly increased AMPK level and insulin content compared to untreated group (p<0.05).
Conclusion: These results support the potential of TP leaf extract as a therapeutic modality for diabetes. However, further studies will be necessary to confirm this potential.

Background: The cytokines have a major role in the inflammatory response to surgery and trauma. They have local effects of mediating and maintaining the inflammatory response to tissue injury, and also initiate some of the systemic changes which occur. IL-6 is one of the above mentioned cytokines, but it is not clear whether the duration of trauma or surgery has some effect on IL-6 level, so the aim of this study was to investigate the relationship between plasma levels IL-6 and duration of orthognathic surgery.
Materials and Methods: Twenty eight patients underwent orthognathic surgery were enrolled in this study. Blood samples were collected one day before and 4 hours after surgery for assessment the levels of plasma IL-6 which was measured by enzyme-linked immunosorbent essay.
Results: We found a significant elevation of plasma IL-6 level at 4 hours after surgery (P<0.05). Moreover, the elevation of plasma IL-6 level was correlated with the duration of surgery.
Conclusion: IL-6 is an important inflammatory mediator that its concentration is increased with the duration of surgery.

The relationship between Human Papillomavirus and Epstein-Barr virus infections with breast cancer of Iranian patients

Background: Breast cancer is the malignancy in humans and other mammals. Several risk factors are involved in their appearance such as higher hormone levels and obesity. Identification of a mouse mammary tumor virus supports a viral etiology for breast tumors in animals. Viruses have been implicated in the development of various cancers, but viral induction for formation breast cancer is controversial. The purpose of this study was investigation of the presence of human papillomavirus (HPV) & Epstein-Barr virus (EBV), in Iranian patients with breast cancer. As the etiology and progression of breast cancer remain incompletely understood, novel routes of disease pathogenesis are important to consider. Some researchers have been found Human papillomavirus (HPV) and Epstein Barr virus (EBV) in breast carcinomas (BCs).
Materials and Methods: Paraffin-embedded sections from 65 female patients with breast carcinoma and 53 breast tissues from patients with fibrocystic disease as control were selected. After DNA extraction and amplification of housekeeping gene (beta-globin), all suitable samples were evaluated for presence of DNA-EBV and DNA-HPV by using PCR (Polymerase Chain Reaction).
Results: HPV DNA sequences were detected in 3 of the 53 benign breast tissue samples but none of the breast carcinoma samples was identified. EBV was detected by PCR in 23 of 65 (35.38%) cases of breast cancer specimens and 11 of 53 (20.75%) control samples ( fibroadenoma 7 of 26 (26.92%) and fibrocystic 4 of 27 (14.81%)). A total of 118 samples from 34 cases (81/28%) were positive. Statistically, crammer indicator analysis for EBV infection in tumor samples and normal samples was 0.46 which indicates a significant relationship.
Conclusion: Our analysis could not confirm a role of HPV in breast cancer but statistically, significant correlation between EBV infection and breast cancer exists. To demonstrate the possible relationship between viral load and breast cancer, need for epidemiological, biological and molecular mechanisms to clear the virus is involved in the process of carcinogenesis.
